A remake had been released before, Super Mario 64 DS (having been released in Japan and North America respectively 2004 and here in Europe 2005). Of course, it knocked my socks off. But let's face it: A second remake with nowadays' engine and some additional exciting feature would be a big hit.
Some fanmade-mods circulating on YouTube and showing what's feasable have knocked my socks off either.
At any rate, it should be really a remake being better than the orignal one and the NDS-remake combined.
Well then, in the following, I'm gonna elaborate the reasons why Super Mario 64 needs a second remake so badly:
1. Famous precedental cases of second remakes of other game-franchises
A famous example for games having been remade twice is the Donkey Kong Country-series (also known in Japan as the Super Donkey Kong-series).
Indeed, the SNES games Donkey Kong Country (1994), Donkey Kong Country 2: Diddy Kong Quest (1995) and Donkey Kong Country 3: Dixie Kong's Double Trouble (1996) received two remakes respectively:

The second time was in the form of Game Boy Advance games having the same titles like the original SNES versions whereat DKC1 having been released in 2003, DKC2 in 2004 and DKC3 in 2005 for the GBA

On the other hand, the GBA-remakes Pokémon FireRed and Pokémon LeafGreen having been released 2004 and on the other hand, the Switch-games Pokémon Let's Go: Eevee and Pokémon Let's Go: Pikachu being released 2018 and based on Pokémon Yellow (1999, in Japan 1998 and in Germany 2000) whereat they additionaly include some elements from Pokémon GO. But by reference to them, you can see what if the Shinys and the regional variants (the Alolan as well as Galarian variants) of certain Pokémon had already occured in Pokémon Yellow and what if that retro-game had been released with the same engine of nowadays Pokémon-games.

Therefore, the just referred game franchises can be considered as precedenctal cases.
For this reason alone, it can be operated with Super Mario 64 to the extent of producing a second remake.
Now, let's transition into the further reason:
2. More characters and features as well as the possibilities by DLCs and patches

Concerning Waluigi, a little excursus: Of course, he deserved a standalone game. When Peach and Toad got respectively a standalone game, why not Waluigi either?
Even Daisy has been impelemted a playable character in Super Smash Bros. Ultimate but unfortunalety, Waluigi had been degraded as Assist Trophy. But there is a Super Smash Bros. Brawl mod being called Super Smash Bros. Legacy XP which is even better than Project M and where you also can play, e.g. as Waluigi. But this is another matter.
Anyways, Waluigi should be a playable character in a second remake of Super Mario 64. Why he hadn't already been dealed in this way in the NDS-remake back then, only Nintendo itself might have the answer.
Furthermore, a Co-Op Mode with max. 4 players might be implemented in the Adventure Mode as well, including the possibility to transfer his lives to a teammate.
But of course, the VS Mode shall also be improved to the extent that not only a free-for-all but also team fight can be selected and that you can play as real Mario, Luigi, Wario and Waluigi and NOT as Yoshi who puts caps in order to take the four former's respective shapes. But not only in the VS.-Mode but also in the Adventure Mode, the transformation hats should be abolished
In order to create more room for the VS.-Mode's expansion, the mini games should better be abolished. Indeed, I liked some of the mini games being included in the NDS-Remake but let us be honest: Nintendo should really get the priorities straight.
Moreover, unlike the NDS-remake, Mario should be playable from the story's very beginning and in the 2nd Bowser Level Bower in the Fire Sea, his youngest son Bowser Jr. (instead of Bowser himself) should be occur as boss villain, in keeping with the motto: "My dad ain't here but a substitution has been provided."
Further features which could also be implemented or alteres are, e.g.:
- The amount of lives should be saved and not be reduced to 5, when you play this game again. Convenient for the possible Co-Op Mode, huh?
- Instead of Yoshi being the one who bestows one-hundred 1UP-Mushrooms on roof of Peach's Castle (like in the original N64-game), it should be another character who could be chosen as NPC in order to do so.
- It should be possible reaching the Castle Courtyard by climbing and passing the hills and roofs similar to the Super Mairo 64-PC-mods

And keep also in mind: Virtually everything is possible with dlcs and patches in order to furtherly improve the game even afterwards so that you don't have to worry about any complaints by the fanbase
3. The implementations of some elements from Super Mario Sunshine

Some of the normal levels of Super Mario Sunshine could also be adapted and improved, let alone, fused.
And Petey Piranha might also be a boss villain in the level Sunshine Isles.
But nevertheless, it should be adapted too much from Super Mario Sunshine because otherwise, it would only go beyond the constraints of a Super Mario 64 remake.
Concluding from all these reasons and aspects mentioned, a second remake from Super Mario 64 for the Switch would indeed be worthwhile precisely because you'd thereby kill several birds with one stone.
